What the researchers found

A standard four-assay toxicology battery (Ames test, in vitro chromosome aberration, acute oral toxicity, bone marrow micronucleus) on hispidin-enriched Sanghuangporus sanghuang mycelia found no statistically significant difference from negative controls, indicating a very low order of toxicity.

Why it matters

This is the most systematic published toxicology assessment for the sanghuang mushroom, and it supplies the preclinical safety floor that the short human trials sit on top of. It also formalises the taxonomic correction: true sanghuang is Sanghuangporus sanghuang, not Phellinus linteus, which matters when transferring safety data between products.

The numbers in context

Hispidin production optimized to 3 mg/g in a 20-ton bioreactor. Four assays: Ames test, in vitro chromosome aberration test, acute oral toxicity test, bone marrow micronucleus test. No statistically significant difference from negative control in any assay.

How the study worked

Culture media optimization across five carbon sources, five nitrogen sources and five initial pH conditions, followed by a genotoxicity and acute oral toxicity battery on the resulting hispidin-enriched mycelia.

Who was studied

In vitro assays and rodent acute oral toxicity / micronucleus testing.

What this study cannot tell us

Tests the mycelium of Sanghuangporus sanghuang, not the fruiting body extract used in the human trials, so the material is not identical to most commercial products. Acute and genotoxic endpoints only, with no chronic or reproductive toxicity assessment. Authors are affiliated with a commercial mycelium producer.
